SUMMARY:Special Seminar - Dr Roberto Bellelli
DESCRIPTION:We are pleased to welcome Dr Roberto Bellelli from The Francis Crick Institute\, to present at CRUK Barts Centre on Tuesday 5th November. \nThe seminar is entitled: ‘Mechanisms of genetic and epigenetic instability: Pol Epsilon and beyond’ \nGenomic instability is a major hallmark of cancer and understanding its nature and origins may provide avenues for patient-tailored therapies. The plasticity of cancer cells is also suggestive of epigenetic mechanisms at the heart of cancer development\, which add a further level of complexity to the challenge of tumour heterogeneity. Indeed\, epigenetic instability\, due to alteration of DNA methylation\, histone post-translational modifications and/or chromatin remodelers has been observed in most malignancies and is now considered a bona fide hallmark of cancer. Despite this\, the interplay between genetic and epigenetic instability remains to be clarified\, particularly in the context of DNA replication\, which represents an opportunity for both genome and epigenome deregulation. \nA paradigm in this area of research is mammalian DNA polymerase Epsilon and its smallest subunits\, POLE3-POLE4\, which I have discovered promotes both efficient replication initiation and facilitates Histone H3-H4 recycling at the replication fork to maintain genetic and epigenetic stability\, respectively.  Through the generation of a mouse knock-out for Pole4 and the identification of POLE1 mutations in patients affected by IMAGe syndrome (Intrauterine growth restriction\, Metaphyseal dysplasia\, Adrenal Hypoplasia and genital anomalies) and immunodeficiency\, I have described the mechanistic consequences of Pol Epsilon instability in genome maintenance\, mammalian development and tumorigenesis. More recently I discovered that POLE3-POLE4 interacts with and chaperones H3-H4 at the replication fork to maintain epigenome integrity and described the molecular determinants of this activity. \nThis work suggests the critical need to dissect in vitro and in vivo the relative contribution of these functions and emphasizes a possible interplay\, yet to be identified\, with histone chaperones\, chromatin remodelers and other replisome components. Identification of these players and their crosstalk in the context of chromatin replication will be of fundamental importance to understand the basis of epigenetic inheritance in mammals and its pathological alteration in cancer. \nThe seminar will be held in the Lister and Young Seminar Room\, 12:30-13:30.

For more information about Richard Beaton’s work please see below. \nThe seminar is entitled: “Sialic acids as innate immune activators; balancing self and non-self to repair or react” \nThe seminar will be held in the Lister and Young Seminar Room\, 12.30pm – 13.30pm. \nRichard is a Research Fellow at King’s College London. Trained in medicine (Glasgow)\, immunology (UCL) and glycobiology (KCL) Richard has been studying the cross-talk between cancer-associated O-linked glycans and host immune cells for over 10 years. He is currently investigating the ability of MUC1 carrying sialylated O-linked glycans to drive monocytes to differentiate into ‘wound healing’ macrophages\, and assessing the prevalence of these processes in breast cancers. Richard has additional interests in gamma delta T cells (for adoptive cancer therapy)\, chronic inflammatory disease (inflammatory arthritis and idiopathic pulmonary fibrosis) and plant derived immunological tools.

DESCRIPTION:We are pleased to welcome Dr Rakesh Heer\, Joint CRUK/RCS(Eng) Clinician Scientist Fellow and hon. Consultant Urological Surgeon – Newcastle University\, to present at Barts Cancer Institute on Monday 11th November.  For more information about Dr Heer’s work\, please click here. \nThe seminar is entitled: ‘Human prostate stem cell biology in homeostasis\, ageing and disease’ \nHaving completed undergraduate medical training in Nottingham in 1997\, Rakesh moved to the North East to undertake surgical training and pursue a career in urology. He became interested in prostate biology and went onto to complete a PhD studying stem cell biology in 2006 at Newcastle University. This work led to two prizes: (i) EUA’s Best Urological Paper (2008) and (ii) a Hunterian Professorship from Royal College of Surgeons of England (2010). This work lay the foundations for a CRUK Clinician Scientist Fellowship that led to a seminal description of human prostate homeostasis. Rakesh was appointed senior lecturer in Newcastle in 2011 pursues specialist clinical and research interests in prostate and bladder cancer. In his talk\, Rakesh will present his work on developing stem cell models to study prostate maintenance\, age-related changes and carcinogenesis. \nThe seminar will be held in the Lister and Young Seminar Room\, 10:00-11:00.

For more information about Dr Läubli’s research\, please click here. \nThe seminar is entitled: ‘Cancer Immunotherapy by Manipulation of Tumor-Associated Glycosylation’. \n\nImpact of the cancer-associated glycosylation on immune suppression\nEnzymatic targeting of cancer-associated glycosylation\nDiscussion of the sialoglycan/Siglec pathway as new target for cancer immunotherapy\nGlycan manipulation for cellular therapies\n\nHeinz Läubli is a physician scientist with clinical training and a specialisation in medical oncology. He is a senior attending physician at the University Hospital in Basel and a head of the laboratory for cancer immunotherapy at the Department of Biomedicine. His research focuses on glycan changes in solid tumors and their influence on the anti-tumor immune response. He is employing genetic and enzymatic manipulation to modify glycans for improvement of cancer immunotherapy.
